The melanocortin angle
MC3R sits primarily in the hypothalamus and is associated with energy homeostasis research. MC4R is involved in appetite regulation, energy expenditure, and reproductive function in preclinical models. PT-141 hits both. Thurston et al. used fMRI to document amplified cerebellar/motor area activity and amygdala-insula functional connectivity during controlled stimulus paradigms in MC4R agonism studies (J Clinical Investigation, 2022; PubMed 36194485).
Structurally, the compound features a norleucine substitution at the N-terminus and a constrained cyclic core via a lactam bridge — giving it defined conformational stability and consistent receptor-binding characteristics in analytical work.
PT-141 vs Melanotan II
Same cyclic core, different receptor profile. Removing the C-terminal amide changes how the peptide interacts with MC3R and MC4R in competitive binding assays. PT-141 research has concentrated on central melanocortin pathway activation, while Melanotan II research covers a broader receptor spectrum.
