MK-677, also known as ibutamoren, is a non-peptide, orally active growth-hormone secretagogue that mimics the action of ghrelin at the GHS-R1a receptor. Unlike the injectable peptides in this category, ibutamoren is a small molecule stable enough for oral delivery, which — together with a research half-life of roughly 24 hours — has made it one of the most studied compounds for investigating sustained activation of the GH/IGF-1 axis.

A non-peptide ghrelin-receptor agonist

Most growth-hormone secretagogues studied in research are peptides that require reconstitution and injection. Ibutamoren is different: it is a spiropiperidine small molecule that binds the ghrelin receptor (GHS-R1a) and, in the models studied, stimulates pulsatile growth-hormone release without suppressing endogenous production. Its oral bioavailability and long duration of action make it a common tool for studying once-daily stimulation of the GH axis.
